Top Three
Scientific Cold Storage Mistakes:
Are your freezers subject to these common
mistakes that can affect the quality of your biologics or compounds? We
are not talking about compressor failures this time. These are mistakes of
normal operation that affect your samples twenty four hours a day:
1) Cycling in Auto-Defrost Freezers.
If you are using automatically defrosting freezers, the
temperature in these units will rise to above 0C at least daily. This
autodefrost period can last for an hour or more every day. Can your
-20C samples handle warm temperatures for these periods. Do your operating
procedures specifically allow temperatures in these kinds of units?
2) Frost. In -80 ultracold
freezers we have measured temperatures that are 40 degrees warmer
than the temperature shown on the front panel. This is seen in
heavily frosted units. Frost acts as insulation. The
temperature shown on the front panel is in a single specific location, and
the temperatures elsewhere in the freezer can be very different.
3) Storage and Measurement Position.
In a upright 2-8C refrigerator there is a gradient of 4 or 5 degrees C
from the top of the unit to the bottom. If your thermometer or other
measurement system is at one point, are you sure that your samples that
are in lower or higher racks are in compliance? These gradients are
even more pronounced in walk-ins and rooms.
